Monoclonal antibodies form an important class of human therapeutics. In the U.S., more than one hundred monoclonal antibodies, including radio-labeled antibodies, bispecific antibodies and antibody drug conjugates (ADC), have been approved by the FDA to date for treatment of cancer, autoimmune diseases, viral and bacterial infection, and other indications. While the immunogenicity associated with murine or other antibodies of non-human origin hampers their utility as therapeutics, humanization technologies make it possible to remove such immunogenicity, allowing clinical use of these antibodies in humans. This has resulted in the development of novel and efficacious therapeutic antibodies, with many of them fulfilling a serious unmet medical need.
JN Biosciences possesses extensive expertise in the humanization of monoclonal antibodies. Using technology described in Tsurushita and Vasquez (Chapter 33, Molecular Biology of B Cells, Elsevier, 2004) and Tsurushita et al. (Methods 2005 36:69-83), JN Biosciences has successfully humanized more than one hundred monoclonal antibodies for external clients or internal programs. Our experience of successful humanization extends beyond murine antibodies and includes antibodies of avian origin.
JN Biosciences offers two levels of humanization services to match the need of our clients. Both are fee-for-service contracts; no future milestones or royalties are requested. The basic level of humanization service provides a client with a design for a humanized antibody based upon client-supplied VH and VL sequences of the antibody of interest. JN Biosciences will use proprietary modeling software and draw upon our extensive experience of successful humanizations to design humanized VH and VL sequences. The second level provides an all-inclusive humanization service. A client simply supplies hybridoma cells (or VH and VL sequences), and JN Biosciences will conduct all of the laboratory work necessary to deliver a quantity, typically several mg, of the purified humanized antibody for evaluation. Customized services are available to meet each client's specific needs.
